Although net uptake of isoleucine and leucine was observed, suggesting that these amino acids were used as oxidative substrate, the increased rate of synthesis appeared to be due to a direct effect on the pathway of protein synthesis rather than an indirect effect mediated by changes in energy levels of the cell.

My understanding about leucine is that it initiates protein synthesis (translation) which means it triggers protein folding and this is the process by which all proteins including muscle protein are made in the body. It starts the process for the body to build various proteins like actin and myosin, proteins which make up muscle tissue. Over loading your body with leucine can have serious consequences as well, unless you are working out regularly and using up these proteins for carbohydrates and ATP. Since the body requires ATP and glucose is the favored source for ATP, our body will use protein to create glucose before it will burn fat. Unless you are consuming a higher than average quantity of protein, you will suffer from muscle wasting if you are trying to burn fat. The RDA for the BCAAs is 50 times lower than what some body building sites suggest. For a female not lifting weights regularly and wanting to weigh about 125 lbs, I figured out 3 grams daily would be enough for me. I eat complex carbohydrates to prevent protein from being catalyzed for glucose. It’s a lot less expensive than consuming protein for glucose via gluconeogenesis although this is the recommended pathway for burning fat and a more frequent protein turnover, although you are using .7 grams of the protein you eat for glucose which would go directly to the brain first then ATP. All I did was calculate how many carbs I would need to consume to stay within the recommended dosages of protein and then from that deduced how many amino acids I would need to meet daily requirements. I do consume protein from food sources as well.

75 grams of protein x .7 = 52.5 gms carbs from protein. 75 – 52.5 = 22.5 gms amino acids. Divide 23 by 8 essentials = ~3gms of each amino.

I have cut my consumption of the BCAAs to 1/2 teaspoon of the GNC product I use, two or three times per day. I get the other essential amino acids from Jo Mar Labs, a small business in Campbell, CA with an office here in Chapel Hill, NC. I have also used Jo Mar Labs BCAAs I like because they are in much lower quantities and equal amounts of each. The risk of getting to much isoleucine is much lower. Isoleucine contributes to blood coagulation. It is also both glucogenic and ketogenic. This is a necessary consideration for smaller bodies.

Not only did I experience sleeplessness after consuming the high dose of BCAAs (10 grams of leucine) but I observed as a result more flabby muscle in the triceps. I wondered if this was from too much glutamate. I was consuming foods high in fat so my thinking was since the BCAAs are hydrophobic, it might also help prevent the clogging of my arteries, pulling the fat into the tissues rather than letting it collect on artery walls. I don’t know if it lowers cholesterol as leucine promotes the production of cholesterol, how much I cannot say.

I will also mention that it has been stated growth hormone is released during sleep. I have read nothing which states the consumption of protein or BCAAs interferes with this release but this might be a consideration since it is the night time fasting which actually triggers the release of HGH.

Again while BCAAs are not synthesized in the liver, according to the article above, nor does their consumption increase energy levels in the muscle cells so overconsumption is of no use to the body and may cause harm. Over consumption of protein can cause other problems such a dehydration, kidney problems and other conditions. There is really no fast track to weight loss or body building. Monitoring food intake, regular exercise and discipline are the key to success. It is advisable to consult a nutritionist or sports nutritionist regarding protein and other dietary needs while dieting or bodybuilding.

A question…

Why does tyrosine zap my appetite? This intrigued me because I typically have very vivid and realistic dreams in the first place. Could melatonin somehow make them more realistic still? Furthermore, if I had the good fortune to recognize such a dream and allow it to become a lucid one, would it be that much better with the help of melatonin?

Erowid Experience Vaults: Melatonin - Interesting Effects on Sleep and Dreaming - 69353
After perusing Erowid I started to wonder if melatonin was similar to LSD or if LSD was a kind of precursor to melatonin in the brain.

My LSD experience was brief and disturbing because I had maintained a tight control on my thinking and behavior for a long time due to traumatic experiences and then was lifted from those experiences and into unfamiliar territory. What I learned was significant and what later unfolded in my personal life was certainly related to that experience. I was not out of control while taking it but my fear kept me from fully grasping and appreciating the reality of what I was experiencing. I can see how people can jump from windows trying to escape something in their mind while doing this substance. It should never be done unsupervised by someone professional. I am guessing it is similar to Prozac as well.

A PCP experience proved fruitful in revealing the content of the mind, but caused me to act out that content rather than see it clearly as the LSD did. Melatonin has permitted me to see, as in remote viewing. No other substance has worked this well for me. I have found there are other things like diet which had an effect on my ability to sleep and stay asleep. Too much protein before bed without glutamine keeps me awake. Too much histamine or too much milk both have an affect on my ability to sleep and in that case I take an anti-histamine like Chlorpheniramine Maleate with the melatonin and/or glutamine. Taking melatonin internally has a powerful effect but under the tongue is much faster. The melatonin/chlorpheniramine combo is better than Abilify. The downside is loss of appetite but since melatonin doesn’t last long in the body this isn’t too much of a problem.

I do not advocate the use of LSD or PCP for these purposes.

ATP and Osteoporosis…

I wonder if osteoporosis is actually caused by adenosine triphosphate since phosphates are part of the bone building process.
After reading this it has become clearer to me that viewing too much television or listening to too much music often prompts the need to take some kind of neuroinhibitory drug such as benzodiazepines or their derivatives. This in turn might affect estrogen levels and cause excessive weight gain even in a young person. Sleeping too much might also cause the problem.

I was given Phenobarbital and Donnatal in high school for acute seizures.
I thought peptides are often actually up-taken faster (than free form aminos) due to specific peptide receptors in the gut. At least this is how it works in general and there may be some aminos that absorb faster than a peptide.

TESTOSTERONE NATION | Amino Acids: Peptide Bond vs. Free Form - Page 1

I have been researching this topic but not with regards to absorption as much as about protein folding. After thinking through the process of protein digestion I came to the conclusion that free form aminos should fold just as any of the amino acids broken down from say a steak after it has been digested.

When I read this response I thought it in error as the issue is absorption so their should be no peptide receptors in the gut related to absorption. Peptides like other proteins have to be broken down further as mentioned in a previous response and then absorbed so actually amino acids would be absorbed faster as they skip this step. Once they are absorbed, then they are folded into their respective conformations as needed by the body and then shuttled off where they are designed to fit into various receptors. I read a sports nutrition book discussing IGF 1 from bovine sources compared to human sources and the author, a sports Doctor, stated the bovine source was not identical to the human source so it was basically not affective. Apparently they were talking about IGF injected into the body rather than ingested via the mouth as even bovine IGF 1 will be broken down into its separate amino acids and then refolded into those needed by the human body. I did read that sometimes these peptides do get through the gut as peptides and can trigger inflammation which would then be attacked by chaperones and ubiquitin, unfolded and then refolded.

I do not use whey anymore because it contains the faster absorbing proteins and this means that the stimulating aminos hit the central nervous system faster and can cause damage as they are absorbed more quickly than casein. I have developed hyperactivity and other symptoms from using whey. I am not a body builder so there is no need for me to consume large quantities of whey.

I have taken amino acids for about 20 years in various forms, even too much of those can cause a problem. I have found them to be more helpful to me and my needs at this time.

I think this is true for humans…

SpringerLink - Neurophysiology, Volume 35, Number 1

…that any excitatory aminos consumed from protein such as whey is going to get converted to glucose rather than getting used by the CNS for neurotransmitters, if no carbs are consumed or low carbs are consumed, particularly of you are an active person. This means fewer excitatory aminos in the central nervous system.

I’ve had a problem getting my body into a state of lipolysis, even to the point of lowering leucine levels so it wouldn’t be stored in adipocytes, like those most found around the abdomen (mostly found on women as protection when pregnant). Unless you are consuming high quantities of protein your body could start breaking down muscle tissue (as mine once did) for carbohydrates as the brain favors carbs for fuel, not enough and brain damage could result. Since lipolysis is a much slower process, carbs via gluconeogenesis will be created much faster which means muscle wasting. Adding large doses of leucine only means more protein synthesis and that could mean breaking down muscle.

I ate lots of fat when I was younger. I didn’t eat lots of protein. I got thin and so did my musculature. The key I found to reducing the brain fog when eating carbs is tyrosine derived from phenylalanine. It keeps you alert and acts like caffeine in the brain but is much better for you. Active people who know anything about their diets know that ATP is important and that carbs are the fastest at creating ATP.

Aside from this, it may be that some species (races) prefer and can utilize a higher fat content versus carbs. I’ve tried it both ways and it seems carbs work for me.

As for seizures, adenosine, the by product of ATP production is neuroinhibitory, preventing seizures. My experience with seizure has been induced either by the excitatory amino acids or by other means outside the body, glutamic acid, glutamate, aspartic acid, alanine and even histidine but not glutamine. A glass of milk can cause me excitation and usually keep me awake if consumed before bed. It should be consumed on an empty stomach. Last night I drank some Coke Zero which contains phenylalanine and aspartic acid. It kept me awake too. I consumed it too late.

Today I consumed 5 grams of phenylalanine along with the other essential aminos (no histidine) in two separate doses as free form amino acids, no aspartic acid. I am not agitated or hyper at all. I have done this frequently for more than 5 years, less frequently for longer in smaller amounts. I also took much less than the recommended dosage for the BCAAs which contains leucine that gets converted to glutamate. Leucine is necessary for protein transcription. It is the levels of circulating essential aminos which contribute to protein folding and the building of proteins which form muscle tissue and other proteins necessary for the functioning of the body.

And I have only one question, if high protein and high fat are good for you, why have heart Doctors and the AHA been telling people to stop eating beef, which can contain as much as 15 grams of saturated fat alone?

The good Doctor also failed to mention the by-product of fat breakdown is glycerol which can pull water from muscles and raise blood pressure. This is what caused Dr. Atkins’ death from a brain aneurysm. Vegetable glycerin from plant foods like celery, cucumbers and zucchini does the same thing. I remember the science class experiment we did in elementary school. A glass of water was filled with food coloring and a stick of celery was placed in the glass of water. Guess what happened? The celery absorbed the colored water.

It seems to me that economics has set the standard for health rather than what is and is not true about the foods we eat.

Who is telling the truth???????

Researchers showed that the spatial learning ability of older rats was impaired, with the impairment correlated to the reduction in taurine in the striatum of the brain (14). Aged rats with modest reductions of taurine showed only modest reductions in learning. Additionally, striatal dopamine was markedly lower in aged learning-impaired rats, demonstrating a potential interaction between taurine and dopamine that may have implications for Parkinson's Disease (see below).

…Unless you are eating nothing but protein and no carbs, eating high amounts of it is going to overexcite your nervous system and even if you do eat it on an empty stomach you are going to experience some excitation. This excitation is what led me to use amino acids. Eating on an empty stomach permits the glucogenic amino acids to be converted into glucose via gluconeogenesis.

This is why there were lawsuits against companies producing aspartame.

I tried a higher protein diet and cutting the carbs but this simply did not work for me and I had already overdosed on glutamic acid once, the other reason I was taking amino acids. It did make sense that eating the protein on an empty stomach would work better but some of the peptides were so readily absorbed and faster acting that I could not use them. I wondered how so few amino acids could possibly be of benefit to good health as stated in the RDA for them and after having read many articles on various amounts of protein needed for adequate health and after having purchased them and taking them according to the RDA, I did not feel I was getting enough protein in my diet and was eating vegetarian. I did not take into account not getting enough fat or carbs. I was getting protein from other sources such as legumes and vegetables. I decided the RDA was not enough.

I will add that taking smaller amounts of the peptide proteins may be better but I still was unable to tolerate then as levels increased during the daily intake. So from my experience, whey would be contraindicated for people with autism and I would venture to say that even milk is a problem for autistics. In taking aminos I would also suggest low levels of phenylalanine which also tends to cause some excitation. Having done all of this, I have come to one conclusion I haven’t read and that is, it seems we may not even need to take in high doses of protein everyday considering all of the other things we consume such as carbs which are derived from protein as cells and some organs do store amino acids. Our bodies will signal us when we need to get more of something in our diets.

Following some of the Atkins recommendations I have put on 50 pounds of fat and now I am having a difficult time getting it off. I reduced my carbs and then my metabolism just slowed to a crawl and I have barely been getting my BMR for calories. Lipolysis takes longer to produce ATP and by the time you get enough your brain and muscle stores could be depleted and you incur brain or muscle damage, not to mention high blood pressure which can lead to aneurysm or other vascular damage. Not only does it cause those problems but one becomes more sedentary in an attempt to preserve muscle stores of glycogen and accommodate the slow production of ATP from fat. While fat may produce more ATP than carbs, it is not the bodies more advantageous source of ATP the energy of life. For one who sits behind a desk all day, a reasonable amount of fat over carbs may be the best remedy for weight gain and satiation, not for active people. My experience in the past with the higher fat, although I was not doing the Atkins it was simply what I was consuming, my body started to breakdown muscle protein to get the carbs I needed in my youth as I was more active and lipolysis was not working. I lost a lot of muscle as a result. Carbs do prevent muscle breakdown. Since some protein does get converted to glucose one could sustain themselves on a higher protein diet if one can tolerate the excitatory amino acids they contain or one finds a source that works better such as casein which contains higher levels of the essential amino acids and lower levels of the excitatory aminos. Milk protein isolate has proven to be of benefit to me and not so stimulating and I have been able to consume it at night with no problems.

Amidst all of this I viewed a film about the giant squid. Thinking about how the squid, over time, develops into a huge creature, it was clear they were doing something right, even though their brains are not highly developed. They only consume the fat from that which they eat from the ocean. They do not eat saturated fats. Their diets are mostly protein and they can get really big without fat. Realizing this I was confused about how Karen Carpenter lost her life because of a high protein diet since more than half a single gram of protein gets converted to carbohydrates in the body, presumably on an empty stomach and leucine is responsible for cholesterol production, not fat, which is necessary in the diet. This leaves one thinking that it was surely the protein that actually caused her death and particularly the excitatory amino acids.

I am sticking to my higher carb diet. It has worked for me in the past so why did I bother changing it? There was a need.

<a href="Brain amino acid requirements and toxicity: the examp... I was never informed of any research being done which may have influenced my actions. I was using GABA to help calm me down after having taken high doses of tyrosine and tryptophan I consumed for a clinical depression, causing my behavior to get out of hand and after having taken too much glutamic acid. I discovered glycine was neuro-inhibitory so I tried it for sleep. It does not aid in sleep even at 3 grams. It does inhibit the frequent motor activity associated with interrupted sleep.

I have found neuro-inhibitory amino acids like glycine can be the cause of manic behavior, especially when there are adequate or high levels of dopamine present. I have taken this amino acid over a period of time. It is neuro-inhibitory and seems to block motor activity when levels are high. When glycine levels are lowered, a rebound affect occurs and causes dopamine to flood synapses, which then causes one to exhibit manic behavior. The same effect can happen when someone takes cocaine that blocks the uptake of dopamine and prevents a sort of resting period after its activation in which dopamine would be taken back up into the cell. Preventing the reuptake can cause cell death from over activation.

My experience with glycine is what led me to conclude the Neanderthal developed his big brain not just from watching and hunting the woolly mammoth but from consuming the woolly mammoth containing high levels of glycine. Neanderthals often consumed the bone marrow consisting of high levels of hydrolyzed protein. Glycine is the most abundant amino acid in hydrolyzed protein. This slower activity enabled the Neanderthal’s brain time to develop before he was assaulted by a barrage of perceived threats to his existence, that would have set into motion all sorts of deadly stress chemicals.

At some point in time during their evolution the woolly mammoth must have consumed foods high in this amino acid. Continued consumption up-regulated the need for this amino acid so they then required more food with which to produce it. As they became vegetarians their levels of other amino acids like dopamine dropped off causing an imbalance in the brain.

The consumption of hydrolyzed protein by humans might have produced developmental problems such as those seen in the Elephant Man. Since my consumption of this amino acid I have developed skin growths resembling those that plagued the body of Joseph Merrick. It may also contribute to Alzheimer’s disease in which case Merrick would have been acting out something from someone else, or acting according to someone else’s will rather than from his own.

<img style="border-right-width: 0px; It grew into a serious problem since seeing The Exorcist, in 1974 which drove me to join the Army. For 6 months I had less than 4 hours of sleep a night. The less sleep I got the more angry I did become over not being allowed to sleep for one reason or another. I felt I had no control of my inability to get adequate sleep and put myself in those situations.

A year or so ago I discovered the film The Woman in Green, which talks about a murder, in which hypnosis was demonstrated to an audience and then to Sherlock Holmes’ sidekick Dr. Watson. It became clear to me, it was possible to be hypnotized by that film and I wondered had I been the victim of my mother having viewed it.

Update 2: The reason I concluded it was this film is because I had purchased a green coat several years prior to discovering this film and green isn’t even my favorite color. I later went back and purchased the same coat in black and gave the green one to my sister, who didn’t need a coat but took it anyway.

Update 3: I was also born at 14 minutes passed midnight so my natural sleep cycle is such that I should be awake at night anyway.

This leads me to think this was the reason I was in a state of hypnotic sleep, but really wide awake physically and acting out the thoughts of the person I was or am connected to in Massachusetts. I am guessing the writer who might have been penning the events resided in Chapel Hill, NC where I had visited in 1972 so the thoughts were still going on in the mind.

Melatonin under the tongue seems to be more affective at inducing sleep…updated

Kay's Science: A good old fashioned cocaine cocktail…

Exogenous ingestion of melatonin and GABA – this makes sense as dreaming and the processing of the days information begins in REM sleep, so it seems melatonin acts like a re-uptake inhibitor, why I am now unable to sleep well again. I am up to 8-10 mgs melatonin at night plus additional glutamine I take in the evening and during the day as well. The darkness in my apartment during the day is causing a problem although I am compensating somewhat with my LCD computer screen set at Standard. This leads one to think our brains must operate more in a manner of gathering information rather than processing it during the daylight ours which is clearly how our more primitive ancestors would have functioned. This explains the keener insights I have been having during the day as my melatonin stores are being released due to my darker apartment. The walls are antique white versus bright white, which I requested as the apartment faces the east and a significant number of trees so it gets little sunshine but lots of heat.

Is this why we have numerous cases of chronic fatigue syndrome? Does melatonin work like a dissociative, as do many cough medicines and why we are so affected by the wake/sleep cycles of others to whom we are psychically connected. This means that during the day we tend not to dissociate unless we have been traumatized by some event of which we are unable to deal, then we slow down and take to our beds for a few days. At night when the sun begins to set [in the case of primitive people their sleep wake cycles are governed more by the rising and setting of the sun] or rather when we decide to lower the lights and start to relax we might start a process of dissociation, whereby we begin to project our daytime rational selves as melatonin is being released into our brains and our bodies become relaxed, catatonic like, and we fall to sleep. This is the reason these chemicals administered exogenously fail over time to promote what they claim as they begin to increase the levels of stimulating chemicals like dopamine despite their other positive effects such as antioxidant activity. The more you take the more you have to take. And, I am guessing, since melatonin has a short half life in the body, about the length of REM sleep, you might awaken after it has been used up, and/or GABA (if levels are adequate) takes over that further increases muscular relaxation, and you then fall into delta sleep. So at once they may act like cocaine but they probably work endogenously in the brain like an orchestra one complementing the other at different times. Therefore one might be better off taking glutamine and tryptophan rather than melatonin and GABA.
Taking it further, this means that with people we are connected to, one might have difficulty sleeping. An example would be having a connection to someone who is a nurse or medical professional who works at night in the bright lights of a hospital. This works fine if you are a day time person and sleep at night. I have experienced this numerous times at my sister’s when visiting and we all are up during the daylight hours. I rarely sleep when I am there, as one of them is always dissociating and I am the recipient of that dissociation. My father experienced this as well despite his blindness although I would guess his melatonin levels depleted after a period of time which affected his ability to sleep at all and became the case. Blind people probably have high levels of melatonin but I am also guessing these deplete unless they consume higher than normal amounts of tryptophan throughout the day.

My experience with consuming tryptophan has been positive and negative. Taken at night tryptophan will induce sleep but after several days of taking it one will become more alert as the sleepy phase wears off and the anti-depressant effects begin to work. The same goes for daytime consumption, there is generally a mild phase of sleepiness followed by greater calm and alertness. It might well be that tryptophan (serotonin) is also causing a degree of dissociation, splitting off the night time drowsiness we experience when we close our eyes and everything becomes dark, so serotonin might be the daytime regulator of melatonin. I have taken it at night frequently and after several days I was simply unable to sleep. I was more energetic and alert. This might all work differently were it not for our psychic connections. I have gone without tryptophan and this makes things work better for me although anorexia becomes an issue as serotonin regulates the hypothalamus which controls appetite, thirst, body temperature, etc. I have also taken a combination of tryptophan, glutamine and vitamin C which was highly effective at helping me sleep.

One evening while viewing the National Geographic video on Sleep Walking, I was struck by the gentleman’s case in which he was charged with the brutal murder of his wife. My first impression was that his neighbor was involved and might have unknowingly affected the man’s behavior. I had been reading and learning about projection and projective identification during that time [who are you?]. As the story unfolded I learned he had difficulties on his job and may have experienced primitive negative thoughts about his boss he could easily have acted out on his wife in a state of sleepwalking. Maybe he was simply having an affair and wanted to get out of his marriage, but why not get a divorce? Why not just leave? Divorcing or leaving are rational ways of dealing with a problem including leaving his job. Fixing a pool pump in the middle of the night is irrational but on a Saturday afternoon would be quite rational.
<div style="padding-bottom: 0px; margin: 0px; padding-left: 0px;
On thinking of this matter in the context of my own life, I recently discovered I might be connected to a woman who is from Chapel Hill. In 1977, after an evening out at a night club, I had a little more to drink than usual and fell asleep on the sofa. I had wet the sofa or bed in my youth and was admonished for it, so I developed repressed memories I expressed as anxiety about falling asleep and the fear of becoming too relaxed in order to keep from wetting the bed and I was a bed wetter until age 12. As a result, that night after drinking too much, I must have had to go to the bathroom, of which I have no recollection, took a white ashtray off the coffee table, placed it on the floor and urinated in it. I did it in this manner because during the late 60’s we had to use porcelain pots to go to the bathroom as we lived in a house with no bathroom for several months. This resolved the mechanics of my actions but not the why of the event. I recently learned my current neighbor who I knew nothing of at the time, during a time of racial unrest, had been stopped from using the public restroom in a local store and was asked to leave. She left the store, got drunk, returned and proceeded to urinate in front of the store. This is as she related her experience to me. She was subsequently arrested by the sheriff (William Blake, sheriff of Orange County, NC, possibly related to my grandmother Eunice Blake) and jailed for one night. I had been jailed overnight in 1977 as well, so this means the drama in which I was involved, had been interrupted, not only was it interrupted, I was completely unconscious of the events occurring in my waking life which led to my behavior that night. I fully believed I was acting of my own will, except for that night. And since this discovery I have located a street, Perly Street, in a small town in Massachusetts with the same last name as my neighbor. I had been in Medford, Massachusetts briefly in 1975 after being discharged from the US Army and so the story goes on further into history, probably pre-Civil War.

What I think about when I start getting confused about which is best, carb or fat, I consider how our brains may have developed. I think about octopi and squid who consumed large amounts of protein and no carbs at all. 0.7 grams of every gram of protein gets converted to carbs. Clearly high protein and low fat is the key to better health and the development of a healthier brain since it was this habit that evolved the human brain to begin with. I just read that manganese must be present for glutamate to be converted into glutamine so it then can be converted to GABA. Minerals the all important key. Taurine, another amino acid derived from methionine, shuttles potassium into the cell and sodium out. I have to agree with the body builders this time that a higher ratio of protein to carbs and fat is the key to better health. The key is the amounts consumed at one time with more frequent consumption. While Karen Carpenter was the extreme of the high protein diet, there is much to be said for it in keeping other conditions under control.

Fat Loss

I have been drinking one cup of coffee in the AM. I have been taking 3-6 mgs melatonin internally and 2-5 mgs under the tongue for the past several months and I am dreaming again. In addition to this I took 2mg chlorpheniramine maleate plus some glycine earlier in the evening. I have been sleeping better than ever. I have also had to reduce the amount of protein I consume in the evenings.

I take the chlorpheniramine maleate because the amino acids I take also contain histidine which can keep one awake. I don’t think it is necessary.

I have also taken this in conjunction with glutamine which does not seem to work as well.

I took three mgs earlier than usual and I was able see things going on in my mind. I thought it was the Abilify but I stopped taking it months ago.

I have also been taking tryptophan, 500 mgs, daily in the AM.

Melatonin instead of GABA might have a powerful effect on the release of growth hormone.
